Description

Cabozantinib is an oral multi-kinase inhibitor that targets several receptor tyrosine kinases involved in tumor growth and angiogenesis. While approved for renal cell carcinoma and hepatocellular carcinoma, cabozantinib is used off-label in colorectal cancer treatment, particularly in patients with specific molecular alterations. The drug shows activity against tumors harboring MET amplifications or RET fusions, which can occur in a subset of colorectal cancers.

Mechanism of Action

Cabozantinib simultaneously inhibits multiple tyrosine kinases including VEGFR2, MET, AXL, RET, and KIT. By blocking these pathways, it disrupts both tumor angiogenesis and direct tumor cell growth signaling, particularly in cancers dependent on MET or RET signaling for proliferation and survival.
