Description
Brivanib is a dual kinase inhibitor that targets both vascular endothelial growth factor receptor (VEGFR) and fibroblast growth factor receptor (FGFR) pathways. In colorectal cancer, brivanib has been evaluated in combination with irinotecan, particularly in patients with elevated plasma FGF levels, and in combination with cetuximab (Erbitux) for advanced gastrointestinal malignancies. The drug was designed to overcome resistance mechanisms that can develop with single-pathway anti-angiogenic therapies by simultaneously blocking both VEGF and FGF signaling.
Mechanism of Action
Brivanib functions as a small molecule tyrosine kinase inhibitor that blocks VEGFR-2 and FGFR-1 receptors, which are critical for tumor angiogenesis and growth factor signaling. By inhibiting both pathways simultaneously, brivanib aims to prevent tumor blood vessel formation through VEGFR blockade while also targeting FGF-mediated resistance mechanisms that can bypass VEGF inhibition.
