Description
BIBF 1120 (nintedanib) is an oral multi-kinase inhibitor that targets angiogenic and fibrotic pathways in cancer treatment. In colorectal cancer, it has been evaluated as an anti-angiogenic agent, potentially serving as an alternative to bevacizumab in metastatic disease. The drug was tested in combination with standard chemotherapy regimens including mFOLFOX6 for patients with advanced, chemotherapy-refractory colorectal cancer.
Mechanism of Action
BIBF 1120 simultaneously inhibits multiple receptor tyrosine kinases including VEGFR (vascular endothelial growth factor receptor), PDGFR (platelet-derived growth factor receptor), and FGFR (fibroblast growth factor receptor). By blocking these angiogenic signaling pathways, the drug disrupts tumor blood vessel formation and maintenance, potentially limiting tumor growth and metastasis.
