Description

BIBF-1120, also known as nintedanib, is a triple angiokinase inhibitor that targets multiple receptor tyrosine kinases involved in angiogenesis and tumor growth. In colorectal cancer, it has been evaluated as an anti-angiogenic therapy, particularly in comparison to bevacizumab for metastatic disease. The drug was tested in combination with standard chemotherapy regimens like mFOLFOX6 for patients with advanced, chemotherapy-refractory colorectal cancer.

Mechanism of Action

BIBF-1120 simultaneously inhibits vascular endothelial growth factor receptors (VEGFR-1, -2, -3), platelet-derived growth factor receptors (PDGFR-? and - ), and fibroblast growth factor receptors (FGFR-1, -2, -3). By blocking these multiple angiogenic pathways, it disrupts tumor blood vessel formation and maintenance, potentially leading to tumor growth inhibition and improved treatment outcomes.
