Description
Axitinib is an oral tyrosine kinase inhibitor that selectively targets vascular endothelial growth factor receptors (VEGFR). In colorectal cancer, axitinib has been evaluated as both monotherapy in chemotherapy-refractory metastatic disease and as maintenance therapy following first-line treatment. The drug has also been studied in combination with standard chemotherapy regimens like FOLFOX and FOLFIRI as an alternative to bevacizumab in metastatic colorectal cancer patients.
Mechanism of Action
Axitinib functions by selectively inhibiting VEGFR-1, VEGFR-2, and VEGFR-3, which are key receptors involved in tumor angiogenesis and vascular development. By blocking these receptors, axitinib disrupts the formation of new blood vessels that tumors require for growth and metastasis.
