Description

Anlotinib is a multi-target tyrosine kinase inhibitor that demonstrates activity in metastatic colorectal cancer as monotherapy and in combination with immunotherapy agents. This oral targeted therapy has shown particular promise in patients with non-liver metastatic disease and as second-line treatment options. Clinical data supports its use both as first-line therapy combined with sintilimab and in second-line settings with irinotecan-based regimens.

Mechanism of Action

Anlotinib functions as a multi-kinase inhibitor that blocks several receptor tyrosine kinases involved in tumor angiogenesis and growth signaling pathways. The drug primarily targets VEGFR1, VEGFR2, VEGFR3, FGFR1-4, PDGFR- / , and c-Kit, thereby inhibiting both tumor vascularization and direct tumor cell proliferation signals.
