Description
Alectinib is an oral anaplastic lymphoma kinase (ALK) tyrosine kinase inhibitor that belongs to the targeted therapy class of anticancer agents. In colorectal cancer, alectinib is utilized for patients whose tumors harbor ALK gene rearrangements, which occur in a small subset of CRC cases. This precision medicine approach targets the specific molecular abnormality driving tumor growth in ALK-positive colorectal cancers. Alectinib represents a tumor-agnostic treatment option for advanced or metastatic solid tumors with confirmed ALK alterations.
Mechanism of Action
Alectinib selectively inhibits the ALK tyrosine kinase receptor and its oncogenic fusion variants that result from chromosomal rearrangements. By blocking ALK signaling pathways, the drug disrupts downstream cellular processes including cell proliferation, survival, and angiogenesis that drive tumor progression. Alectinib also demonstrates activity against the RET tyrosine kinase receptor, providing additional antitumor effects.
