Description
Adagrasib is a small molecule KRAS G12C inhibitor specifically designed to target tumors harboring the KRAS G12C mutation in colorectal cancer. This targeted therapy works by covalently binding to the mutant KRAS G12C protein, effectively blocking downstream signaling pathways that drive tumor growth. Adagrasib is used in metastatic colorectal cancer patients whose tumors test positive for the KRAS G12C mutation, often in combination with cetuximab and other agents.
Mechanism of Action
Adagrasib functions as a covalent inhibitor that specifically targets the KRAS G12C mutant protein by binding to the cysteine residue unique to this mutation. By locking KRAS G12C in its inactive GDP-bound state, adagrasib prevents the protein from activating critical downstream signaling pathways including RAF-MEK-ERK and PI3K-AKT that promote cancer cell proliferation and survival.
